چکیده

In the course of a study on the osidation of various aldehydes by glyceraldehyde 3-phosphate dehydrogenase (l), it was observed that aged preparations of glycolaldehyde 2-phosphate inhibited strongly the activity of glyceraldehyde Q-phosphate dehydrogenase. Differences in inhibitory potency of several orders of magnitude were noted between various batches. A synthetic preparation, kindly supplied by Dr. C. E. Ballou, had no effect even at relatively high concentrations; however, a solution of the same preparation after several months of storage contained a potent inhibitor. A study of the conditions leading to the formation of inhibitor, revealed that it was rapidly formed from glycolaldehyde 2-phosphate in 1 N NaOH at room temperature. Isolation and analysis of the inhibitor showed it to be a tetrose diphosphate. It is the purpose of this paper to report on the preparation of the tetrose diphosphate and on the mechanism of its inhibitory action on glyceraldehyde 3-phosphate dehydrogenase.

New reactive coenzyme analogues for affinity labeling of NAD+ and NADP+ dependent dehydrogenases.

Reactive coenzyme analogues omega-(3-diazoniumpyridinium)alkyl adenosine diphosphate were prepared by reaction of omega-(3-aminopyridinium)alkyl adenosine diphosphate with nitrous acid. In these compounds the nicotinamide ribose is substituted by hydrocarbon chains of varied lengths (n-ethyl to n-pentyl). The diazonium compounds are very unstable and decompose rapidly at room temperature. They ...

A Labile C02-Fixing Enzyme Complex in Spinach Chloroplasts

Activity and activation of ribulose 5-phosphate kinase, ribulose diphosphate carboxylase, phosphoglycerate kinase, and the NADPand NAD-dependent glyceraldehyde phosphate dehydrogenases from isolated chloroplasts were traced in tbe course of differential ultracentrifugation. Activation of some of these enzymes was carried out by incubation with NADPH2 or ATP. Activity of ribulose diphosphate car...

NAD+-dependent glyceraldehyde-3-phosphate dehydrogenase from Thermoproteus tenax. The first identified archaeal member of the aldehyde dehydrogenase superfamily is a glycolytic enzyme with unusual regulatory properties.

The hyperthermophilic archaeum Thermoproteus tenax possesses two glyceraldehyde-3-phosphate dehydrogenases differing in cosubstrate specificity and phosphate dependence of the catalyzed reaction. NAD+-dependent glyceraldehyde-3-phosphate dehydrogenase catalyzes the phosphate-independent irreversible oxidation of D-glyceraldehyde 3-phosphate to 3-phosphoglycerate. The coding gene was cloned, seq...

H2O2-induced block of glycolysis as an active ADP-ribosylation reaction protecting cells from apoptosis.

H2O2 treatment on U937 cells leads to the block of glycolytic flux and the inactivation of glyceraldehyde-3-phosphate-dehydrogenase by a posttranslational modification (possibly ADP-ribosylation). Glycolysis spontaneously reactivates after 2 h of recovery from oxidative stress; thereafter cells begin to undergo apoptosis. The specific ADP-ribosylation inhibitor 3-aminobenzamide inhibits the str...
